Depends on the tire.
On my TKC-80 tires, the little triangular marks just point at the wear
indicators.
To verify whether a particular mark is the "heavy" or "light" side of
the tire, I look inside the tire for the seam where the carcass halves
overlap. This will be a thicker part. That's the heavy part of the
tire. You want to put that opposite the stem. Then I look at where the
mark is at relative to that, and use the mark to properly position the
tire while I'm installing it (hard to look inside the tire when the
rim is halfway down on it
