Stock engine. Stock jetted carb. with shimmed needle & idle mixture screw out 1.75 turns.
If I do a tweety-ectomy, how bad of a hit will my mpg take?

I get between 42 and 47 mpg in mostly city riding in Los Angeles, depending on how hard I ride, with the exact same configuration.

I have been getting 50 mpg from day one. I have a 09 and commute 75
miles per day at a speeds around 75 on open road. Thinking about
changing the gearing to a 16 tooth for a better top end. Maybe this
will increase my Mpg.

The 16T is a tradeoff. I ran it for a while (about 5,000 miles) and went back to the 15T. If you try it, I'd be interested to see if your mileage might actually go DOWN, because you end up holding the throttle open wider to maintain freeway speeds.
If you do it, keep us posted.
Here are my impressions:
Plus:
Freeway cruising @75MPH felt more "relaxed" (about 400RPM down from where the 15T operates).
Each gear pulls a little longer.
Minus:
You lose a LOT of the torque that we all crave. Much more than I expected.
*Takeoffs in 1st require a little more clutch action
*Passing torque (git up 'n' go) is abysmal (60-80MPH runs, etc)
*Top-end isn't much higher, if any. With the 15T your top speed is limited by redline, with the 16T it will not reach redline in 5th.
*High Winds and steeper hills require downshifts.
*First gear is a little too fast for hairy trail riding.
The worst gas mileage I ever got on my KLR was !!34MPG!! Northbound on I-5. I was fighting a 35MPH headwind. With the 16T, the bike would BARELY maintain 60MPH with the throttle pinned in 5th.
I switched back and have been happier with having the "grunt" back.
But, hey... It only costs $20.00 to try, so there's little to lose.

I've got an 06 I bought used last year...It had a Dyna Jet Stage 2 kit
http://klr650.com/jetkit.htm and K&N air filter and a Jardine RT-4 Canister muffler
http://klr650.com/Jardine.htm when I bought it ...I added the Quiet Module insert to muffle the noise a bit more and I'm running a 14 tooth countersprocket and Metzler Karoo II's....I ride It where ever I want to (highway, street ,dirt) and have always got 55 MPG ...I also run Royal Purple synthetic oil ...I don't know why it gets such good mileage but this seems to be a good combo and It works for me.

I ran a 16T on a trip to Alaska and got really bad mileage. It was badly overgeared, could barely get up some of hills, had to run the hell out of it, no power. Switched back to a 15T when I got back home and the mileage went way back up. Didn't change anything else...same panniers and all. The Rev told me not to go to a 16 and I did it anyway...he know what he was talking about. A loaded KLR with a 16T is a real pig, in my experience...I got 36 mpg on that trip, and normally get 44-46 with the stock sprockets. Not too good.

I changed to a 16t and haven't seen a change in my mileage. I am usually
loaded lightly though. I still feel like I have plenty of power on the
freeway too. It keeps my rpm below 5000 usually which eliminates any oil
burning.

cam and balancer chains?

Anybody ever replace their chains (cam and Balancer)? A friend says
he is going to at 50,000 miles. Mine just turned 52,000. We were
checking my doohickey spring as he checked his after 20,00 after
replacement with new doohickey and it was broke. Mine was OK ......
but one of the washers on the starter gear was missing??????
